ATI-0810 (formerly PG301029)
ATI-0810 has been found to inhibit the replication of hepatitis C virus by interfering with late stage replication events involved with RNA and protein production. Mechanistic evaluations suggest that the compound inhibits HCV replication by a unique mechanism which does not involve inhibition of virus entry, polymerase, protease or IRES functions. Thus, ATI-0810 represents a novel inhibitor of HCV replication and will be an entirely new class of HCV therapeutics upon introduction into human clinical trials in 2009. Additionally, ATI-0810 appears to have broad based inhibitory potential which includes HCV, HBV and HIV, acting in each case through a transcriptional/RNA synthesis mechanism of inhibitory action. Interestingly, combination studies have shown that the addition of ATI-0810 to the clinically approved combination of ribavirin and interferon results in a synergistic increase in the efficacy of the combination drug therapy. Cell based assays indicate that the synergy is due in part to a reduction in the toxicity of ribavirin which is observed upon addition of ATI-0810 to the therapy. A pre-IND data package has been submitted to the FDA in anticipation of a human clinical trial for the product in 2009.
